Common genes and pathways involved in prediabetes and diabetes using bioinformatics analysis

Diabetes mellitus is a global, potentially fatal metabolic disorder affecting millions, with predictions of a doubling of affected individuals within two decades [1]. MicroRNAs (miRNAs) are crucial in diabetes mellitus, regulating gene expression and affecting insulin release, β-cell proliferation, and function. Dysregulated miRNAs can lead to insulin resistance and impaired glucose homeostasis [1,2]. In this study, using the GEO database, GSE26468, which had normal, diabetes, and pre-diabetes samples, was analyzed. First, the diabetic and pre-diabetic groups were compared with the normal group and the differentially express genes (DEGs) was calculated. The Enrichr was used for enrichment pathway analysis. Then common DEGs in diabetes and pre-diabetes were identified. STRING was used to get the protein- protein (PPI) network. The PPI was analyzed with Gephi software and the most important proteins with the highest degree were selected. Related miRNAs were obtained from miRTarBase database. The regulatory network was reconstructed with Cytoscape software. Enrichment pathway analysis ans biological process showed insulin signaling pathway, growth hormone signaling pathway and BCR signaling pathway and regulation of apoptotic process, mucosal immune response, integrin-mediated signaling pathway involved in prediabetes and diabetes. Also, TPM3, GRB2, BCL2L1, PAK2, miR-34a-5p, miR-15b-5p and miR-124-3p have regulatory role in miRNA-mRNA interaction network. If experimental studies confirm our results, such studies can provide valuable insights to understand the role of miRNAs in the pathogenesis of diabetes, to develop targeted interventions and improve patient outcomes.
