Definition of SARS-CoV-2 proteins antigenicity by computational immunology

Introduction: COVID-19 caused by SARS-COV-2 (severe acute respiratory syndrome coronavirus 2) might lead to severe respiratory problems need intensive care (Heyman, 2020). From September 2021, millions of people have been infected by COVID-19 and some died around the world. Although some vaccines against SARS-COV-2, have been effective in COVID-19 mortality control (Ji and Wang, 2020), so far no exact safe and high effective vaccine has been offered. Identification of SARS-CoV-2 proteins with high antigenicity for more effective vaccines development could be a priority (Zhu and Zhang, 2020). In present study, SARS-CoV-2 proteins antigenicity was determined by computational immunology. Methods: Proteins of SARS-CoV-2 comprising S, M, E, N and ORF3a, with an essential role in virus virulence and reproduction, were selected. Then the amino acid sequences of these proteins were attained in NCBI database at www.ncbi.nlm.nih.gov in FASTA setup. Afterward, antigenicity of proteins was evaluated by Vaxigen V.2 software. Proteins with score > 0.4 were selected as antigen. Results: All SARS-CoV-2 proteins evaluated in this study had antigenic score > 0.4. Thus these proteins could be considered as antigen. Conclusions: According to results of this study S, M, E, N and ORF3a proteins of SARS-CoV-2 are antigens and so could stimulate the immune system. For preparation of more effective anti- SARS-COV-2 vaccines, additional studies aimed at recognition the epitopes of S, M, E, N and ORF3a proteins with higher immunogenicity which could induce vigorous immune response are required.
