Immunoinformatics Approach to Epitope-Based Vaccine Design for H5N1 Avian Influenza Virus

The H5N1 avian influenza virus poses a significant global health threat, with the potential to cause pandemics and high mortality rates. Despite efforts, effective vaccines remain limited. This study explores the design of an epitope-based vaccine for H5N1, utilizing advanced tools for predicting T-cell and B-cell epitopes to develop targeted immune responses and improve vaccine efficacy. We first extract Zika virus epitopes from the IEDB site.then Extracted epitopes were predicted by ToxinPred, AllerTop,Vaxijen servers in terms of allergenicity, toxicity,and immunogenicity.Then the confirmed MHC 1 epitopes were connected with AAY linker.Then,MHC2 was connected to the previous ones with the GPGPG linker, and finally,the epitopes of B cells were connected with the KK linker.To increase the immunogenic power, Adjuvant should be added to the structure,and from the NCBI site,the sequence of interleukin- 12,partial [Rattus sp.] was added to the structure of epitopes.Finally,His-Tag, i.e. 6 H numbers,was added to purify the vaccine.We rechecked our structure with ToxinPred, AllerTop,Vaxijen servers to ensure that it is not allergenic,non-toxic,and has a favorable immunogenicity. Then we checked the designed vaccine and its molecular weight was 45659.95 daltons and also the isoelectronic point of our protein was 8.92.It was also in a stable state and the Aliphatic index was 68.71.Then we checked the second structure with the jalview server,the result was favorable and also the third structure was checked with phyre2 server and it had 99.1% confidence with c7purB.
